Quit all of your apps and/or reboot... check to see if your cam works in a different app (like iChat or iMovie).
It'll help to determine if it's the camera or just the software having a hiccup.

It looks like you have an iChat session going and photo booth won't open simultaneously? Just a guess, I'm still getting use to my new system. But that symbol means you don't have your camera on (green light by the built-in) like grim said.
